I applied online. The process took 1+ week. I interviewed at Lowe's Home Improvement in Mar 2008
Interview
You fill out an online background form. They invite you in for a personality test and faace-to-face interviews with at least 2 managers. If you pass the drug test, they will make you an offer based on current openings. Most of the timie they offer a part-time position. Take it, because they usually convert the part timers they like to full time after a few months.

Interview
Was contacted 3 days after applying and told to come in for a interview 2 days later. Went there and met with the HR manager had initial interview with her then met with department manager then store manager. Some questions asked caught me off guard like what would you describe your greatest weakness to be, other then that it was typically easy and the managers I met were nice they gave me a drug test which i had to hold a stick in my mouth then the interview was over they called me few days later and told me when to come in and start. The pay was better then i had before but not negotiable.

Interview
HR manager interview, then interviewed with the store manager. Pretty basic questionaire that both are reading from and scoring you right in front of you. Not too stressful because you can tell they aren't really interested in what you're saying. Totally depends on who you end up interviewing with though, I've worked in a few different stores and the level of professionalism among managers varies greatly.
